No, the 12th Captain Underpants book is not the last one in the series. So far there have been 13 books released in the series, with the most recent being Captain Underpants and the Sensational Saga of Sir Stinks-A-Lot.
The original plan of creator Dav Pilkey was to have 12 books in the series, but that was eventually extended to an additional book in 2015. There have been no plans to release any more books in the series as of yet, but Pilkey has not ruled out the possibility.

We can safely expect new books to be released in the series at some point in the future, as it’s not just a favorite among children and young adults, but also among adults.
